A method is disclosed for inducing cell-mediated immunity against cellular antigens. More specifically, the invention provides for a method for inducing cytotoxic T-lymphocyte immunity against weak antigens, notably self-proteins. The method entails that antigen presenting cells are induced to present at least one CTL epitope of the weak antigen and at the same time presenting at least one foreign T-helper lymphocyte epitope. In a preferred embodiment, the antigen is a cancer specific antigen, e.g. PSM, Her2, or FGF8b. The method can be exercised by using traditional polypeptide vaccination, but also by using live attenuated vaccines or nucleic acid vaccination. The invention furthermore provides immunogenic analogues of PSM, Her2 and FGF8b, as well as nucleic acid molecules encoding these analogues. Also vectors and transformed cells are disclosed. The invention also provides for a method for identification of immunogenic analogues of weak or non-immunogenic antigens.
On décrit un procédé qui permet d'induire l'immunité à médiation cellulaire contre des antigènes cellulaires. Plus spécifiquement, il s'agit d'un procédé permettant d'induire l'immunité des lymphocytes T cytotoxiques contre les antigènes faibles, plus particulièrement les protéines autologues. Le procédé implique que les cellules présentatrices de l'antigène sont incitées à présenter dans le même temps au moins un déterminant antigénique CTL de l'antigène faible et au moins un déterminant antigénique de lymphocyte T auxiliaire étranger. Dans une forme de réalisation préférée, l'antigène est un antigène spécifique contre le cancer, par exemple PSM, Her2 ou FGF8b. Le procédé peut être mis en oeuvre au moyen de la vaccination polypeptidique classique, mais également au moyen de vaccins vivants atténués ou par vaccination d'acide nucléique. L'invention concerne également des analogues immunogènes de PSM, Her2 et FGF8b, ainsi que des molécules d'acide nucléique codant ces analogues. Des vecteurs et des cellules transformées sont présentés. L'invention concerne en outre un procédé d'identification d'analogues immunogènes d'antigènes faibles ou non immunogènes.
