Methods for generating chimeric polynucleotides by directed evolution are described. In the methods, splice points of interest are identified within the polynucleotides of a basis set of polynucleotides, preferably through the use of an algorithm that defines the number of splice points and selects the splice points, either by random selection or using information regarding alignment of the polynucleotides. The algorithms can include additional factors, including a definition of a desired distance between splice points, and/or weighing factors to bias selection of splice points. Chimeric polynucleotides are generated using primers (e.g., double primers or non- overlapping primers) and polymerase chain reaction or combinatorial strategies.

L'invention concerne des méthodes permettant de générer des polynucléotides chimériques par évolution dirigée. Dans ces méthodes, des points d'épissage étudiés sont identifiés dans les polynucléotides d'un ensemble de base de polynucléotides, de préférence, par utilisation d'un algorithme définissant le nombre de points d'épissage et sélectionnant les points d'épissage, soit par sélection aléatoire soit au moyen d'informations relatives à l'alignement des polynucléotides. Les algorithmes peuvent comprendre des facteurs supplémentaires, notamment une définition d'une distance souhaitée entre des points d'épissage et/ou des facteurs de pondération permettant d'orienter la sélection des points d'épissage. Des polynucléotides chimériques sont générés au moyen d'amorces (par exemple des amorces doubles ou des amorces non chevauchantes) et d'une réaction en chaîne par la polymérase ou de stratégies combinatoires.
