Abstract A process for preparing 3.alpha.,7.alpha.-dihydroxy-12-keto-5.beta.- cholanic acid is disclosed comprising: a bacterium having productivity of 3.alpha.,7.alpha.-dihydroxy-12-keto-5.beta.-cholanic acid belonging to genus Bacillus is cultivated in a nutrient medium containing 3.alpha.,7.alpha.,12.alpha.-trihydroxy-5.beta.-cholanic acid, wherein pH of said medium is preferably 7 to 11, and the product in the culture medium is collected. Said bacterium is a mutant strain of Bacillus, and which has productivity of said acid with substantially 100% conversion and substantially 100% yield.
