An isolated c-Fos regulating kinase polypeptide (FRK) characterized by having a molecular weight of 88 kD as determined by reducing SDS-PAGE, having threonine and serine kinase activity, phosphorylating the c-Fos activation domain and polynucleotide sequences and method of detection of FRK are provided herein. Also described are methods for identifying compositions which affect FRK activity, thereby affecting c-Fos activation and subsequent activation of genes associated with AP-1 sites.
L'invention porte sur un polypeptide isolé de kinase de régulation de c-Fos (FRK) caractérisé par un poids moléculaire de 88 kD déterminé par électrophorèse sur gel polyacrylamide en présence de dodécyl sulfate de sodium (SDS-PAGE) réducteur, exerçant une activité de kinase de thréonine et de sérine, phosphorylant le domaine d'activation de c-Fos, et sur des séquences de polynucléotides et une méthode de détection de FRK. L'invention porte également sur des méthodes d'identification de compositions affectant l'activité de FRK et, en conséquence, affectant l'activation de c-Fos et, subséquemment, l'activation de gènes associés à des sites AP-1.
