A protein which inhibits osteoclast differentiation and/or maturation and a method of production of the protein. The protein is produced by human embryonic lung fibroblasts and has molecular weight of about 60 kD and about 120 kD under non-reducing conditions and about 60 kD under reducing conditions on SDS-polyacrylamide gel electrophoresis, respectively. The protein can be isolated and purified from culture medium of the said fibroblasts. Furthermore, the protein can be produced by gene engineering. The present invention includes cDNA for producing the protein by gene engineering, antibodies having specific affinity to the protein or a method for determination of the protein concentration using the antibodies.
La présente invention concerne une protéine nouvelle, qui a pour effet de supprimer la différenciation et/ou la maturation des ostéoclastes. Elle concerne aussi des procédés de production de cette protéine. La protéine en question est produite à partir de fibroblastes pulmonaires de foetus humains. Son poids moléculaire est d'environ 60 KD dans des conditions réductrices et d'environ 120 KD dans des conditions non réductrices. Elle peut être isolée et purifiée en partant du milieu de culture des cellules mentionnées. Elle peut aussi être produite au moyen de techniques du génie génétique. L'invention concerne aussi un ADNc pour la production de la protéine par génie génétique, un anticorps qui manifeste une affinité spécifique pour cette protéine et un procédé de dosage de celle-ci utilisant cet anticorps.
