The invention concerns novel compositions and methods for controlling nucleic acid expression in cells. More particularly, it concerns any nucleic acid characterised in that it comprises: a) a first region comprising a nucleic acid coding for a tetracycline (tTA)-dependent transactivator under the control of a moderate promoter; and b) a second region comprising a nucleic acid of interest under the control of tTA sensitive promoter. The invention is more particularly useful for controlling the expression of nucleic acids in the nerve cells, in vitro as well as in vivo, for example the human tyrosine hydrocyclosis.

La présente invention concerne de nouvelles compositions et méthodes pour contrôler l'expression d'acides nucléiques dans les cellules. Elle concerne en particulier tout acide nucléique caractérisé en ce qu'il comprend: a) une première région comprenant un acide nucléique codant pour le transactivateur du système de régulation par la tétracyline (tTA) sous contrôle d'un promoteur modéré, et, b) une deuxième région comprenant un acide nucléique d'intérêt sous contrôle d'un promoteur sensible au tTA. L'invention est plus particulièrement adaptée au contrôle de l'expression d'acides nucléiques dans les cellules nerveuses, in vitro comme in vivo, par example de la tyrosine hydroxylase humaine.
