Compositions for the diagnosis and therapy of prostate and colon cancer, derived from or based on a novel prostate-specific, androgen-regulated, cell surface serine protease termed 20P1F12/TMPRSS2 are described. A full length cDNA comprising the entire coding sequence of the 20P1F12/TMPRSS2 gene (also designated 20P1F12-GTC1 herein) is provided (FIG. 1). Among the compositions provided are antibodies that bind to 20P1F12/TMPRSS2 proteins and polypeptide fragments thereof, including antibodies labelled with a detectable marker or toxin or therapeutic composition. Several monoclonal antibodies specifically reactive with 20P1F12/TMPRSS2 are also described herein.
L'invention concerne des compositions, utiles pour le diagnostic et la thérapie propres au cancer de la prostate et du colon, qui sont dérivées d'une nouvelle sérine protéase de surface cellulaire à régulation par androgène et spécifique à la prostate dénommée 20P1F12/TMPRSS2, ou bien qui reposent sur cette protéase. L'invention concerne en outre de l'ADNc entier comprenant la totalité de la séquence de codage du gène 20P1F12/TMPRSS2 (également dénommé 20P1F12-GTC1 aux fins de la présente invention). Parmi les compositions décrites figurent des anticorps qui se lient aux protéines 20P1F12/TMPRSS2 et à des fragments polypeptidiques de ces protéines, y compris les anticorps porteurs d'un marqueur, d'une toxine ou d'une composition thérapeutique détectable. L'invention concerne enfin plusieurs anticorps monoclonaux réagissant spécifiquement à 20P1F12/TMPRSS2.
