Methods for identifying modulators of nuclear hormone receptor function comprise the steps of (a) forming a mixture comprising a nuclear hormone receptor, a peptide sensor and a candidate agent, but not a natural coactivator protein of the receptor, wherein the sensor provides direct, in vitro binding to the receptor under assay conditions; (b) measuring an agent-biased binding of the sensor to the receptor, and (c) comparing the agent biased binding with a corresponding unbiased binding of the sensor to the receptor. In particular embodiments, the sensor comprises an amphipathic alpha helix nuclear hormone interacting domain comprising a recited nuclear hormone transcriptional coactivator motif sequence, the sensor is present at sub- micromolar concentration, the binding reaction occurs in solution, the sensor comprises a fluorescent label and the measuring step comprises detecting fluorescence polarization of the label. Reagents include labeled sensor peptides and reaction mixtures consisting essentially of nuclear hormone receptor, a peptide and a candidatent.
L'invention porte sur des procédés d'identification de modulateurs de la fonction des récepteurs de l'hormone nucléaire, ces procédés consistant à: (a) obtenir un mélange comprenant un récepteur de l'hormone nucléaire, un capteur peptidique et un agent candidat, et non une protéine coactivatrice naturelle du récepteur, le capteur assurant la liaison directe, in vitro, au récepteur dans des conditions de dosage; (b) mesurer une liaison sollicitée par un agent du capteur au récepteur; et (c) comparer la liaison sollicitée par un agent avec une liaison correspondante non sollicitée du capteur au récepteur. Selon des réalisation spécifiques, le capteur comprend une hormone nucléaire à hélice alpha, amphipathique, ayant une interaction avec un domaine comprenant une séquence à motifs structuraux coactivatrive, transcriptionnelle de l'hormone nucléaire précitée; ou bien le capteur est présent dans une concentration submicromolaire, ou la réaction de la liaison a lieu dans la solution, ou encore le capteur comprend une étiquette fluorescente, l'étape de mesure consistant à détecter la polarisation de la fluorescence de l'étiquette. Selon une autre réalisation, des réactifs comprennent des peptides de capteurs marqués et des mélanges réactionnels comprenant essentiellement un récepteur d'hormone nucléaire, un peptide et un candidat.
