The present invention provides labeled nucleic acid amplification oligonucleotides, which can be linear or hairpin primers or blocking oligonucleotides. The oligonucleotides of the invention are labeled with donor and/or acceptor moieties of molecular energy transfer pairs. The moieties can be fluorophores, such that fluorescent energy emitted by the donor is absorbed by the acceptor. The acceptor may be a fluorophore that fluoresces at a wavelength different from the donor moiety, or it may be a quencher. The oligonucleotides of the invention are configured so that a donor moiety and an acceptor moiety are incorporated into the amplification product. The invention also provides methods and kits for directly detecting amplification products employing the nucleic acid amplification primers. When labeled linear primers are used, treatment with exonuclease or by using specific temperature eliminates the need for separation of unincorporated primers. This "closed- tube" format greatly reduces the possibility of carryover contamination with amplification products, provides for high throughput of samples, and may be totally automated.
La présente invention concerne des oligonucléotides marqués qui amplifient des acides nucléiques. Ces oligonucléotides peuvent être linéaires, d'amorce en U ou de blocage. Les oligonucléotides de l'invention sont marqués avec des fractions donneuses et/ou receveuses de paires de transfert d'énergie moléculaire. Les fractions peuvent être des fluorogènes, auquel cas l'énergie fluorescente émise par la fraction donneuse est absorbée par la fraction receveuse. La fraction receveuse peut être un fluorogène émettant à une longueur d'ondes différente de celle de la fraction donneuse, ou un extincteur de luminescence. Les oligonucléotides de l'invention sont configurés de telle sorte qu'une fraction donneuse et une fraction receveuse sont incorporées dans le produit d'amplification. L'invention concerne aussi des procédés et des trousses permettant une détection directe des produits d'amplification utilisés avec les amorces d'amplification de l'acide nucléique. Lorsque des amorces linéaires marquées sont utilisées, un traitement par un exonucléase ou à une température spécifique supprime la nécessité de séparer des amorces non incorporées. Ce format en "tube fermé" limite considérablement la possibilité d'une contamination de report par les produits d'amplification, fournit des débits d'alimentation élevés d'échantillons et peut être entièrement automatisé.
