The present invention provides a nitrile hydratase nucleic acid fragment isolated from Pseudomonas putida which encodes a nitrile hydratase activity capable of catalyzing the hydrolysis of certain racemic nitriles to the corresponding R- or S- amides. Also provided are transformed microorganisms capable of the active expression of said nitrile hydratase activity. Additionally, the invention provides a transformant harboring the nitrile hydratase gene in conjuction with an amidase gene, both of which may be co- expressed producing active nitrile hydratase and amidase enzymes respectively. Methods for the production of such enantiomeric materials are also provided.
Fragments d'acide nucléique de nitrile hydratase isolés à partir de Pseudomonas putida, codant une activité nitrile hydratase capable de catalyser l'hydrolyse de certains nitriles racémiques vers les R- ou S- amides correspondants. L'invention porte également sur des micro-organismes transformés capable d'exprimer activement ladite activité nitrile hydratase. De plus, l'invention porte sur un transformant abritant le gène nitrile hydratase en conjonction avec un gène amidase, l'un et l'autre pouvant être co-exprimé et donner lieu respectivement à une nitrile hydratase active et à des enzymes amidases. L'invention porte enfin sur des méthodes de production de tels matériaux énantiomères.
