The invention concerns a nucleic acid capable of regulating the ABCA7 gene transcription, which codes for a carrier protein capable of intervening in the metabolism of lipids and/or in the process involving the immune system and inflammation. In addition, owing to the position of the gene on the chromosome 19 in q 13, ABCA7 is potentially involved in other pathologies genetically related to said locus. The invention also concerns nucleotide constructs comprising a polynucleotide coding for a polypeptide or a nucleic acid of interest, placed under the control of a nucleic acid regulating the ABCA7 gene. The invention further concerns recombinant vectors, transformed host cells and non-human transgenic mammals comprising a nucleic acid regulating the ABCA7 gene transcription and or said nucleotide construct, and methods for screening molecules or substances capable of modulating the activity of the nucleic acid regulating the ABCA7 gene.
La présente invention concerne un acide nucléique capable de réguler la transcription de gène ABCA7, qui code pour une protéine transporteur susceptible de jouer un rôle dans le métabolisme des lipides et/ou dans les processus impliquant le système immunitaire et l'inflammation. Par ailleurs, de par la position du gène sur le chromosome 19 en q13, ABCA7 est potentiellement impliqué dans d'autres pathologies liées génétiquement à ce locus. La présente invention est également relative à des constructions nucléotidiques comprenant un polynucléotide codant pour un polypeptide ou un acide nucléique d'intérêt, placé sous le contrôle d'un acide nucléique régulateur du gène ABCA7. L'invention a également trait à des vecteurs recombinants, des cellules hôtes transformées et des mammifères transgéniques non humains comprenant un acide nucléique régulateur de la transcription du gène ABCA7 ou une construction nucléotidique précitée, ainsi que des procédés pour le criblage de molécules ou de substances capables de moduler l'activité de l'acide nucléique régulateur de gène ABCA7.
