A restriction site indexing method for selectively amplifying any fragment generated by a Class II restriction enzyme includes adaptors specific to fragment ends containing adaptor indexing sequences complementary to fragment indexing sequences near the termini of fragments generated by Class II enzyme cleavage. A method for combinatorial indexing facilitates amplification of restriction fragments whose sequence is not known. Profiling methods and other methods for characterizing polynucleotides are presented.
L'invention concerne un procédé de classification de site de restriction, permettant d'amplifier sélectivement tout fragment produit par une enzyme de restriction de classe II. Ce procédé utilise des adaptateurs spécifiques aux extrémités desdits fragments, dont les séquences de classification d'adaptateur sont complémentaires aux séquences de classification de fragment, à proximité des parties terminales des fragments produits par un clivage enzymatique de classe II. L'invention concerne également un procédé de classification combinatoire facilitant l'amplification de fragments de restriction, dont la séquence n'est pas illustrée. L'invention concerne enfin des procédés permettant d'établir le profil des polynucléotides, ainsi que d'autres procédés destinés à caractériser ces polynucléotides.
