The invention relates to a nucleic acid molecule, containing a nucleic acid which codes for a polypeptide with chorismate mutase activity and derivatives thereof, whereby the derivatives have at least 10 % of the chorismate mutase activity of the chorismate mutase, according to the identification number of the SEQ ID NO:2. The invention further relates to vectors containing nucleic acid molecules, to host cells comprising nucleic acid molecules and their use in methods for producing polypeptides with chorismate mutase activity. The invention also relates to the polypeptides with chorismate mutase activity and antibodies which specifically recognise the same. In addition, the invention relates to methods for producing auxotrophic yeast strains using the nucleic acid molecules and to the preparation of the yeast strains. The invention also relates to the use of yeast strains together with the inventive vectors in methods for the recombinant expression of heterologous genes which allow the transformants to be easily selected by functional complementation.
La présente invention concerne une molécule d'acide nucléique contenant un acide nucléique codant pour un polypeptide à activité de chorismatmutase et pour des dérivés de celui-ci, ces dérivés présentant au moins 10 % de l'activité de la chorismatmutase correspondant au numéro d'identification de séquence SEQ ID NO:2. L'invention concerne en outre des vecteurs contenant des molécules d'acide nucléique, des cellules hôtes comprenant des molécules d'acide nucléique et leur utilisation dans des procédés de production de polypeptides à activité de chorismatmutase. De plus, l'invention concerne lesdits polypeptides ayant une activité de chorismatmutase et des anticorps reconnaissant spécifiquement ceux-ci. L'invention concerne également des procédés de production de souches de levure auxotrophes à l'aide desdites molécules d'acide nucléique, ainsi que la préparation des souches de levure. L'invention concerne de surcroît l'utilisation des cellules souches avec des vecteurs selon l'invention, dans des procédés d'expression, par recombinaison, de gènes hétérologues qui permettent d'effectuer facilement la sélection des transformants par complémentation fonctionnelle.
