The present invention is directed to nucleic acids encoding novel truncated CART polypeptides, particularly CART proteins having 88 amino acids in length, as well as nucleic acid fragments thereof, the related recombinant vectors and recombinant host cells containing such recombinant vectors. The invention also deals with a truncated CART protein particularly a truncated CART protein having 88 amino acids in length, uses thereof in the identification of ligands, as well as antibodies specially directed against this protein, these antibodies being useful as diagnostic reagents. The invention also relates to the use of the nucleic acids encoding the truncated CART proteins of the invention or the truncated CART proteins themselves for therapeutic purposes and particularly to decrease or prevent neuronal degeneration and to promote neuronal regeneration and restoration of function, to treat addiction by aiding clearance of the polypeptide or its truncated forms in vitro and in vivo and also in order to regulate food intake.
L'invention concerne des acides nucléiques codant pour des nouveaux polypeptides CART tronqués, en particulier des protéines CART d'une longueur de 88 acides aminés ainsi que les fragments d'acide nucléique de ces derniers. L'invention concerne également une famille de vecteurs recombinants qui contiennent un acide nucléique codant un tel polypeptide CART tronqué et des cellules hôte recombinantes qui contiennent de tels vecteurs recombinants. L'invention concerne par ailleurs une protéine CART tronquée, en particulier une protéine CART tronquée comptant 88 acides aminés en longueur ainsi que des anticorps spécifiquement dirigés contre cette protéine, ces anticorps étant utiles en tant que réactifs de diagnostic. L'invention concerne en outre l'utilisation d'une protéine CART, en particulier d'une protéine CART tronquée possédant 88 acides aminés en longueur ou de fragments de celle ci pour l'identification et la caractérisation des ligands qui se lient à la protéine CART ou à son/ses récepteur(s). L'invention concerne enfin l'utilisation d'acides nucléiques codant pour ces protéines CART tronquées ou des protéines CART elles-mêmes à des fins thérapeutiques, en particulier pour réduire ou prévenir la dégénérescence neuronale et pour stimuler la régénération neuronale et la restauration des fonctions, pour traiter la toxicomanie en favorisant la clairance du polypeptide ou de ses formes tronquées in vitro et in vivo, ainsi que pour réguler de l'ingestion d'aliments.
