The invention relates to double-stranded nucleic acid fragments comprising a chemically modified backdone and at least 4-1000 bp, preferably 8-500 bp, and most preferably 16-200 bp. The disclosed molecules (DRIL molecules) may interfere with DNA damage signalling and repair pathways, in particular the non homologous NHEJ pathway of double-stranded break repair. The invention discloses the application of the DRIL molecules as adjuvant compositions to be used in association with a DNA breaking treatment, particularly radiotherapy or chemotherapy, in combination with a pharmaceutically acceptable carrier, in an efficient amount to be introduced in the tumoral cell nuclei in order to trigger DNA repair induced lethality (DRIL in short) of tumoral cells/tissues.

L'invention concerne des fragments d'acide nucléique bicaténaire comprenant un squelette modifié chimiquement et au moins 4-1000 bp, de préférence 8-500 bp et idéalement 16-200 bp. Les molécules selon l'invention (molécules DRIL) peuvent s'immiscer dans les voies de signalisation et de réparation du dommage à l'ADN damage, notamment la voie non homologue NHEJ de réparation des cassures bicaténaires. L'invention concerne l'application de molécules DRIL en tant que compositions adjuvantes destinées à être utilisées en association avec un traitement de rupture de l'ADN, notamment une radiothérapie ou une chimiothérapie, combiné à un excipient acceptable sur le plan pharmaceutique, à un taux efficace pour être introduit dans les noyaux des cellules tumorales afin de déclencher la létalité induite par la réparation de l'ADN (DRIL) des cellules ou tissus tumoraux.
