The present invention relates to the discovery, identification and characterization of a novel subgroup of activins/inhibins, herein referred to as liver activins, that during vertebrate adult life are specifically expressed in the liver. The invention relates to the activin .beta.C and .beta.E subunits and other members of the liver activin subgroup. The invention encompasses liver activin nucleotides; liver activin genomic regulatory elements that regulate the expression of liver activins; host cell expression systems; liver activin proteins, fusion proteins, polypeptides and peptides; antibodies to liver activin protein, transgenic animals that express a liver activin; recombinant knock-out animals that do not express liver activin(s); antagonists and agonists of the liver activins; and other compounds that modulate liver activin gene expression activity to regulate cell growth and/or differentiation and to diagnose and treat abnormalities related thereto. In addition, the present invention encompasses methods and compositions for the regulation of cell growth and/or differentiation including, but not limited to, stimulating liver regeneration, bone growth and hematopoiesis.
La présente invention a trait à la découverte, l'identification et la caractérisation d'un nouveau sous-groupe d'activines/inhibines, ci-après désignées sous le nom d'activines du foie, spécifiquement exprimées dans le foie chez un vertébré adulte. L'invention concerne des nucléotides d'activines du foie; des éléments régulateurs génomiques d'activines du foie régulant l'expression d'activines du foie; des systèmes d'expression de cellules hôtes; des protéines d'activines du foie, des protéines de fusion, des polypeptides et des peptides; des anticorps contre une protéine activine du foie, des animaux transgéniques exprimant une activine du foie; des animaux recombinés dont le gène inactivé ("knock out") n'exprime pas une (des) activine(s) du foie; des antagonistes et des agonistes des activines du foie; et d'autres composés modulant l'activité d'expression du gène d'activine du foie afin de réguler une croissance et/ou une différenciation cellulaire et de diagnostiquer et traiter des anomalies y relatives. En outre, la présente invention concerne également des procédés et compositions permettant la régulation d'une croissance et/ou la différenciation cellulaire, y compris, de façon non exhaustive, la stimulation d'une régénération du foie, d'une croissance osseuse et d'une hématopoïèse.
