Nucleotide sequences encoding the variable heavy and light chains of the murine monoclonal antibody 1F7 are disclosed. The 1F7 antibody is an effective immune modulator that has anti-idiotypic binding affinity for anti-HIV antibodies. Polypeptides containing at least one complementarity-determining region (CDR) or framework-determining region (FR) of the variable heavy or variable light chains of 1F7, as well as the polynucleotides encoding them, can be used to modulate the immune response to HIV infection. Unnecessary, and potentially adverse, murine segments of the variable chains falling outside the CDRs can be replaced with human sequences to afford humanized chimeric antibodies and antibody fragments.
L'invention concerne des séquences nucléotidiques codant les chaînes légères et lourdes variables de l'anticorps 1F7 monoclonal murin. L'anticorps 1F7 est un immunomodulateur efficace qui présente une affinité de liaison anti-idiotypique pour des anticorps anti-VIH. Des polypeptides contenant au moins une région de détermination de complémentarité (CDR) ou une région de détermination de cadre (FR) des chaînes légères variables ou lourdes variables de l'1F7, ainsi que les polynucléotides les codant, peuvent être utilisés pour moduler la réponse immunitaire à l'infection VIH. Des segments murins non nécessaires, et potentiellement hostiles, des chaînes variables tombant en dehors des CDR peuvent être remplacés par des séquences humaines pour fournir des anticorps chimériques humanisés et des fragments d'anticorps.
