Nucleotide sequences have been isolated that encode a C16 specific ACP thioesterase. The instant nucleotide sequences are expressed in E. coli and plant tissue. These sequences have been used in the anti-sense inhibition of endogenous plant thioesterase and in the regulation of the aryl co-enzyme A pool for the reduction of saturated fatty acid content in vegetable oil.
