2064692 9102086 PCTABS00003 A method of assaying a plurality of soil samples collected at intervals across an exploration territory for subsurface oil or gas deposits, which method comprises: (i) determining from a first portion of a first sample a first value which represents a first microbial activity and which is attributable to microorganisms which metabolise a selected hydrocarbon gas; (ii) determining from a second portion of the first sample a second value which represents a second microbial activity and which is attributable to a representative proportion of all microorganisms in the sample; (iii) repeating steps (i) and (ii) for further samples; and (iv) determining for each sample the first microbial activity relative to the second microbial activity from the said first and second values obtained for each sample. The concentration of free hydrocarbon gas may also be taken at each site at which a soil sample is taken and combined with the determination made in step (iv).
