A method for the separation of a target molecule from a mixture is described. The method employs oil bodies and their associated proteins as affinity matrices for the selective, non-covalent binding of desired target molecules. The oil body proteins may be genetically fused to a ligand having specificity for the desired target molecule. Native oil body proteins can also be used in conjunction with an oil body protein specific ligand such as an antibody or an oil body binding protein. The method allows the separation and recovery of the desired target molecules due to the difference in densities between oil bodies and aqueous solutions.

L'invention concerne un procédé permettant d'isoler une molécule cible à partir d'un mélange. Le procédé fait appel à des corps huileux et à leurs protéines associées comme matrices d'affinité pour la fixation sélective non covalente des molécules cibles désirées. Les protéines des corps huileux peuvent être génétiquement fusionnées à un ligand présentant une spécificité pour la molécule cible désirée. Les protéines natives des corps huileux peuvent également être utilisées en conjonction avec un ligand spécifique d'une protéine de corps huileux, tel qu'un anticorps ou une protéine de fixation de corps huileux. Le procédé permet de séparer et de récupérer les molécules cibles désirées grâce à la différence de densité entre les corps huileux et les solutions aqueuses.
