Oligonucleotide tagged nucleoside triphosphates, OTNTPs, which are substrates for polymerases and or terminal nucleotidyl transferases are provided as well as methods of making these OTNTPs. Further provided are OTNTPs with fluorescent dyes including energy transfer dyes, attached to the oligonucleotide chain, OTNTPs with unnatural bases incorporated in the oligonucleotide sequence and methods for incorporating these OTNTPs in DNA or RNA. Also provided are methods for using the oligonucleotides on OTNTPs for amplifying the oligo sequence on the OTNTP using an amplification method described above. Further provided are reactive bifunctional amidites, methods of making these compounds and methods for detecting single nucleotide polymorphisms using the above OTNTPs. Methods for detecting differential gene expression using the OTNTPs and methods of separating specifically modified DNA or RNA using the OTNTPs are also provided.
L'invention concerne des triphosphates nucléosidiques marqués par oligonucléotides OTNTP, lesquels sont des substrats pour des polymérases et/ou des nucléotidyles transférases terminales, ainsi que des procédés de production de ces OTNTP. L'invention concerne également des OTNTP à colorants fluorescents, notamment des colorants à transfert d'énergie, fixés à la chaîne oligonucléotidique, des OTNTP ayant des bases non naturelles contenues dans la séquence oligonucléotidique, et des procédés d'incorporation de ces OTNTP dans l'ADN ou l'ARN. L'invention concerne également des procédés d'utilisation des oligonucléotides dans les OTNTP pour amplifier la séquence oligo dans l'OTNTP à l'aide d'un procédé d'amplification. L'invention concerne aussi des amidites réactives bifonctionnelles, des procédés de production de ces composés ainsi que des procédés de détection de polymorphismes mononucléotidiques à l'aide des OTNTP précités. L'invention concerne aussi des procédés de détection de l'expression différentielle de gènes à l'aide des OTNTP ainsi que des procédés de séparation d'ADN ou d'ARN modifiés spécifiquement à l'aide des OTNTP.
