Oligonucleotide comprising, from its 5' end to its 3' end, a first portion comprising an upstream segment and a downstream segment forming a pin-loop structure, and a second portion not capable of self-pairing, in which the first portion comprises at least one blocking agent capable of blocking the replication, by a polymerase, of the said oligonucleotide, such that at least a portion of the downstream segment is replicated and the upstream segment is neither totally nor partially replicated. This oligonucleotide may be used as a primer for the cyclic amplification of a target sequence of a nucleic acid.
Oligonucléotide comprenant, de 5' vers 3', une première partie comprenant un segment amont et un segment aval formant une structure tige-boucle, et une seconde partie non susceptible d'auto-appariement, dans lequel la première partie comprend au moins un agent bloquant, capable de bloquer la réplication par une polymérase dudit oligonucléotide, de façon qu'au moins une partie du segment aval soit répliquée et de façon que le segment amont ne soit répliqué ni totalement ni partiellement. Cet oligonucléotide est utilisable comme amorce dans un procédé d'amplification cyclique d'une séquence cible d'un acide nucléique.
