A new selective, rapid and highly sensitive method (as few as 2 × 10 -15 mole) for the detection of human .alpha.-thrombin is disclosed, based on the formation of hybrid anionic aptamer/cationic polythiophene complexes. Advantageously, this new method does not require any chemical reaction on the probes or the analytes and is based on conformational modifications of the conjugated backbone of a cationic poly(3-alkoxy-4-methylthiophene), when mixed with ss-DNA and a target protein.
