The invention is related to a nucleic acid molecule comprising a polynucleotide encoding a modified filovirus glycoprotein (GP) having at least one amino acid change located in a relatively conserved region of said GP that decreases in vitro cytotoxicity and retains immunogenicity when compared to in vitrocytotoxicity and immunogenicity of a wild type filovirus GP, and related modified filovirus GPs, plasmid DNAs, recombinant viruses, adenoviruses, pharmaceutical compositions, vaccine compositions, antibodies that are specifically reactive with the modified filovirus GPs, and related methods of making and using the same.
L'invention porte sur une molécule d'acide nucléique comprenant un polynucléotide codant une glycoprotéine (GP) de filovirus modifiée possédant au moins une variante d'acide aminé située dans une région relativement conservée de la glycoprotéine qui réduit la cytotoxicité in vitro et conserve une immunogénicité comparée à la cytotoxicité in vitro et l'immunogénicité d'une glycoprotéine de filovirus de type sauvage. L'invention porte également sur des glycoprotéines de filovirus modifiées apparentées, des ADN plasmidiques, des virus de recombinaison, des adénovirus, des compositions pharmaceutiques, des compositions vaccinales, des anticorps qui réagissent spécifiquement aux glycoprotéines de filovirus modifiées, et sur leurs procédés de fabrication et d'utilisation.
