A process is provided for preparing an oral fast-melt composition of a selective cyclooxygenase-2 inhibitory drug comprising (a) a step of wet granulating the selective cycloorxygenase-2 inhibitory drug together with a binding agent selected from grums, polypetpides, natural and modified starches, cellulosic materials, alginic acid and salts thereof, polyethylene glycol, polyvinylpyrrolidone, polymethacrylates, silicate salts and bentonites, and (b) a step of blending with the drug a saccharide of low moldability, wherein the above steps (a) and (b) occur in any order or simultaneously to result in formation of granules. Optionally the process further comprises (c) a step of blending the granules with at least one of a lubricant, a sweetening agent and a flavoring agent to form a tableting blend, and (d) a step of compressing the tableting blend to form oral fast-melt tablets. Also provided is a composition prepared by such a process.
L'invention concerne un procédé de préparation d'une composition orale à dissolution rapide d'un médicament sélectif d'inhibition de la cyclooxygénase-2. Ce procédé comprend les étapes consistant (a) à effectuer une granulation par voie humide dudit médicament conjointement avec un agent de liaison sélectionné dans des gommes, des polypeptides, des amidons naturels et modifiés, des matières cellulosiques, de l'acide alginique et des sels de celui-ci, du polyéthylène glycol, du polyvinylpyrrolidone, des polyméthacrylates, des sels de silicate et des bentonites et (b) à mélanger un saccharide de faible aptitude au moulage avec le médicament, les étapes (a) et (b) étant effectuées dans un ordre quelconque ou de façon simultanée, de manière à obtenir une formation de granulés. Le procédé peut, éventuellement, comprendre des étapes consistant à (c) mélanger les granulés avec au moins un lubrifiant, un agent adoucissant et un agent aromatisant, de manière à former un mélange destiné à la fabrication de comprimés, et (d) à comprimer ledit mélange, de manière à former des comprimés oraux à dissolution rapide. L'invention concerne également une composition préparée au moyen d'un tel procédé.
