An osmotic delivery system for controlled delivery of a beneficial agent includes an implantable capsule containing a beneficial agent and an osmotic engine that swells on contact with water, to thereby cause the release of the beneficial agent over time. The osmotic delivery system has a membrane material that allows a controlled amount of fluid to enter from an exterior of the capsule, while preventing the compositions within the capsule from passing out of the capsule. The osmotic delivery system is designed to meet at least the operating pressures of 1000 psi. the membrane material is cast, calendered or extruded followed by machining (i.e. die-cutting, stamping or otherwise cutting to shape) to provide a uniform non-ribbed membrane material. The capsule also includes a membrane material retaining means that is positioned at a fluid uptake end to retain the membrane material within the capsule, even under periods of high pressure.

L'invention concerne un système d'administration osmotique destiné à une administration régulée d'un agent thérapeutique et comprenant une capsule implantable contenant un agent thérapeutique et un moteur osmotique se gonflant au contact de l'eau, ce qui provoque la libération progressive de l'agent thérapeutique. Ce système d'administration osmotique comporte une matière membranaire permettant l'entrée d'une dose régulée de fluide à partir d'une zone extérieure de la capsule tout en empêchant les compositions de la capsule de passer à l'extérieur de cette capsule. Ledit système d'administration osmotique est conçu pour atteindre au moins des pressions d'utilisation de 1000 psi. La matière membranaire est coulée, calandrée ou extrudée avant de subir un usinage (et notamment un découpage à l'emporte-pièce, un estampage ou un autre formage par coupage), d'où l'obtention d'une matière membranaire uniforme non nervurée. La capsule comprend également un organe de retenue de matière membranaire situé au niveau d'une extrémité d'absorption de fluide en vue de retenir la matière membranaire à l'intérieur de la capsule, y compris pendant des périodes de haute pression.
