The gene for outer membrane protein P1 of Haemophilus influenzae b is expressed in E. coli. Methods for expression and demonstration of the immunogenicity of recombinant P1 and portions thereof are disclosed, along with an improved method for the purification of P1. The nucleotide sequence of the P1 gene and the derived amino acid sequence of the P1 protein of Haemonhilus influenzae type b are disclosed and the methods used to determine the same. Also disclosed are the methods used to clone and express the P1 gene as wall as the purification protocol for the P1 gene products (recombinant P1 and P1 fusion proteins). Fourteen peptides are synthesized corresponding to specific sequences of the mature P1 protein. The use of the P1 protein as n immunogens for immunization against the disease caused by Haemophilus influenzae type b and the use of the protein as a carrier for conjugation with an oligosaccharide derived from Haemophilus to generate a potentially efficacious vaccine against the disease, are described. Also disclosed is the use of P1 peptide-conjugates as immunizing agents to elicit anti-Haemophilus influenzae type b antibodies.
Le gène produisant la protéine de membrane externe P1 de l'Haemophilus influenzae de type b est exprimé dans l'E. coli. Des procédés servant à l'expression et à la manifestation de l'immunogénicité de la protéine P1 recombinante et de parties de cette protéine, ainsi qu'un procédé amélioré permettant la purification de la protéine P1 sont décrits. La séquence de nucléotides du gène de la protéine P1 et la séquence d'acides aminés dérivée de la protéine P1 de l'Haemophilus influenzae de type b ainsi que les procédés utilisés pour déterminer ces séquences sont décrits. Les procédés utilisés pour le clonage et l'expression du gène de la protéine P1 ainsi que le protocole de purification des produits géniques de la protéine P1 (Protéine P1 recombinante et protéines de fusion de la protéine P1) sont également décrits. Dans ces procédés, on effectue la synthèse de 14 peptides qui correspondent à des séquences spécifiques de la protéine P1 mûre. L'utilisation de la protéine P1 sous la forme de n immunogènes assurant une immunisation contre la maladie causée par l'Haemophilus influenzae de type b et l'utilisation de la protéine comme véhicule permettant sa conjugaison avec un oligosaccharide dérivé de l'Haemophilus en vue de produire un vaccin potentiellement efficace contre cette maladie, sont en outre décrites. L'utilisation de conjugués peptidiques de la protéine P1 comme agents d'immunisation entraînant la production d'anticorps anti-Haemophilus influenzae type b est également décrite.
