Method and constructs are provided for the synthesis of a new and useful single-stranded DNAs which have a stem-loop configuration (slDNA). The process of the prior art were dictated and limited by the endogenous level of primer that initiated replication from the origin of replication of a recombinant DNA vehicle. In accordance, new and useful methods and constructs for a system wherein a set of genetic elements that control the replication of the DNA vehicle operate independently of the set of genetic elements which control slDNA synthesis. Further, the initiation of slDNA synthesis is not coupled to or depended upon the replication of the recombinant DNA vehicle.
