A mutein of an enzyme acceptor polypeptide fragment of .beta.-galactosidase which is resistant to oxidation is provided. The enzyme acceptor fragment has an amino acid other than cysteine located at position 602 of the natural sequence. An especially preferred substitution is serine. Also provided are a method for providing the novel mutein, a reagent composition comprising the novel mutein, and an immunoassay method for determining an analyte in which the novel mutein recombines with an enzyme donor polypeptide fragment to form enzymatically active-.beta.-galactosidase.

Mutéine d'un fragment polypeptidique accepteur d'enzyme de la .beta.-galactosidase, résistant à l'oxydation. Le fragment accepteur d'enzyme a un acide aminé autre que la cystéine dans la position 602 de la séquence naturelle. Un acide de substitution préféré est la sérine. Il est également prévu un procédé de production de la nouvelle mutéine, une composition réactive contenant la nouvelle mutéine, et un procédé d'analyse immunologique permettant de déterminer une substance à analyser, dans lequel la nouvelle mutéine se recombine avec un fragment polypeptidique donneur d'enzyme pour former de la .beta.-galactosidase active du point de vue enzymatique.
