A variety of genetic constructs are disclosed that will find both in vitro and in vivo use in the area of tumor biology an cancer therapy. In particular, expression construcs are provided that contain a p16 encoding region and other regulatory elements necessary for the expession of a p16 transcript. One version of the expression construct is a replication-deficient adenoviral vector. Also provided are methods for the transformation of cell lines and the inhibition of cancer cell proliferation.

L'invention se rapporte à une série de produits de synthèse génétiques susceptibles d'être utilisés in vitro et in vivo dans les domaines de la biologie tumorale et de la thérapie anticancéreuse. On décrit en particulier des produits de synthèse à expression qui comportent une région de codage de p16 et d'autres éléments régulateurs nécessaires à l'expression d'un produit de transcription de p16. Une version du produit de synthèse à expression se présente sous la forme d'un vecteur adénoviral à réplication déficiente. On décrit par ailleurs des procédés applicables à la transformation des lignées cellulaires et à l'inhibition de la prolifération des cellules cancéreuses.
