In the absence of substantial sequence overlap between a recombinant adenoviral vector and the genome of a packaging cell, helper dependent El- containing particles (HDEP) can be formed at low frequency. The invention provides means and methods reducing or preventing the generation of HDEP. To this purpose, novel packaging cells and methods of making these are provided.

En l'absence de recouvrement substantiel de séquences entre un vecteur d'adénovirus de recombinaison et du génome d'une cellule d'encapsidation, on peut former à faible fréquence des HDEP (particules à dépendance de cellules T auxiliaires contenant des E1). L'invention porte sur des moyens et des méthodes de réduction ou prévention de la création d'HDPE, et sur de nouvelles cellules d'encapsidation et leurs méthodes d'élaboration.
