The invention relates to the detection of a polynucleotide using a padlock probe, whereby a hybridization process is combined with ligation to irreversibly capture polynucleotides of interest. The invention relates both to the padlock probe itself, as well as to methods of use, which allow for varying levels of sensitivity due to amplification of signal processes built into the system. Additionally, preferred formats are disclosed for use of the padlock probes, including the use of microfluidics.

La présente invention se rapporte à un procédé de détection d'un polynucléotide au moyen d'une sonde cadenas, dans lequel on combine un processus d'hybridation et une ligature pour capturer de manière irréversible des polynucléotides spécifiques. L'invention se rapporte à la fois à la sonde cadenas elle-même ainsi qu'à des procédés d'utilisation, qui permettent d'obtenir des degrés divers de sensibilité grâce à l'amplification des processus générateurs de signaux élaborés au sein du système. L'invention se rapporte en outre à des formats préférés d'utilisation de ces sondes cadenas, et notamment à l'utilisation de microfluides.
