The present invention relates generally to growth factors and more particularly to growth factors which are capable of stimulating or otherwise facilitating formation of insulin-secreting cells. The identification of these growth factors permits the development of protocols to culture cells in vitro for transplantation into mammalian and in particular human subjects with insulin-dependent type 1 diabetes or related conditions. It is further contemplated that the endogenous expression of growth factors required for the development of insulin-producing cells may be manipulated in vivo, by the appropriate administration of agents including genetic agents capable of regulating the expression of growth factors in pancreatic duct epithelial cells. The growth factors may also be administered to subjects with type 1 diabetes to stimulate the proliferation and differentiation of pancreatic cells into insulin-secreting cells. The present invention also provides modulators of growth factor-mediated pancreatic cell differentiation. Such modulators are useful in the treatment inter alia of .beta. cell tumors and/or pancreatic cancer.
La présente invention porte de façon générale sur des facteurs de croissance et plus précisément sur des facteurs de croissance capables de stimuler ou de faciliter d'une autre manière la formation de cellules insulino-sécrétrices. L'identification de ces facteurs de croissance permet le développement de protocoles servant à cultiver des cellules in vitro en vue d'une transplantation chez un mammifère, en particulier chez des sujets humains présentant des diabètes du type 1 insulino-dépendants ou des états pathologiques associés. Selon la présente invention, l'expression endogène des facteurs de croissance requis pour le développement des cellules productrices d'insuline peut être manipulée in vivo par l'administration appropriée d'agents, notamment des agents génétiques capables de réguler l'expression des facteurs de croissance dans des cellules épithéliales du canal pancréatique. Les facteurs de croissance peuvent être également administrés aux sujets présentant des diabètes du type 1 en vue de stimuler la prolifération et la différentiation cellulaires pancréatiques dans des cellules insulino-sécrétrices. La présente invention porte également sur des modulateurs de différentiation cellulaire pancréatique induite par les facteurs de croissance. Ces modulateurs sont utiles dans le traitement inter alia des tumeurs de la cellule .beta. et ou du cancer du pancréas.
