A method of analyzing a polynucleotide of interest, comprising providing one or more sets of consecutive oligonucleotide primers differing within each set by one base at the growing end therof; annealing a single strand of the polynucleotide or a fragment of the polynucleotide to the oligonucleotide primers under hybridization conditions; subjecting the primers to single base extension reactions with a polymerase and terminating nucleotides, the terminating nucleotides being mutually distinguishable; and observing the location and identity of each terminating nucleotide to thereby analyze the sequence or a part of the nucleotide sequence of the polynucleotide of interest, is disclosed. An apparatus comprising a solid support to which is attached at defined locations thereon one or more sets of consecutive oligonucleotide primers differing within each set by one base at the growing end thereof is also described.
Procédé d'analyse d'un polynucléotide à étudier, qui consiste à obtenir un ou plusieurs ensembles d'amorces consécutives d'oligonucléotides, qui diffèrent au sein de chaque ensemble par une base située à la terminaison de croissance de ladite amorce, à circulariser un brin simple du polynucléotide ou d'un fragment du polynucléotide aux amorces d'oligonucléotides dans des conditions d'hybridation, à soumetttre les amorces à des réactions d'extension de base unique à l'aide d'une polymérase et de nucléotides de terminaison, lesdits nucléotides de terminaison pouvant être mutuellement distingués, et à observer l'emplacement et l'identité de chaque nucléotide de terminaison pour analyser ainsi la séquence ou une partie de la séquence nucléotidique du polynucléotide étudié. Un appareil comportant un support solide auquel sont fixées en deux emplacements définis une ou plusieurs séries d'amorces consécutives d'oligonucléotides qui diffèrent au sein de chaque ensemble par une base située à la terminaison de croissance de ladite amorce est également décrit.
