ABSTRACT OF THE DISCLOSURE A diaphragm pump cassette for use with a pumping mechanism. The cassette comprises a pumping chamber and a check valve assembly communicating with the pumping chamber. The check valve assembly includes a first check valve and a second check valve. Each check valve includes a valve seat, flexible valve diaphragm and a movable valve seating mechanism for locating the diaphragm on the valve seat. The valve diaphragms are unseated with respect to the valve seat prior to mounting the pump cassette on the pumping mechanism whereby fluid may flow freely through the check valve. The valve seating mechanism of the first check valve includes a further mechanism, independent of the pumping mechanism, for manually opening and closing the first check valve.
