2082262 9215675 PCTABS00016 A method for the particle-mediated transformation of cotton permits the direct genetic engineering of elite cotton lines without the need for tissue culture or callus proliferation. The technique involves excising the embryonic axes from germinating seeds and blasting particles carrying foreign genes into the embryonic axes. When the treated axes are released from an initial cytokinin treatment, the meristems recover and generate a shoot. By using a marker gene, the transformant population of the shoots can readily be identified. In this fashion, direct and expeditious transformations of elite cotton lines in Pima, Acala, Sea Island and Upland varieties can routinely be achieved.
