The present invention relates to peptides and peptidomimetic compounds of formula (I) and pharmaceutical compositions containing them as useful pharmacological agents in the control or treatment of proliferative diseases such as cancer, against tumor growing and/or tumoral metastasis, and psoriasis and in the control or treatment of inflammatory, allergic, autoimmune, viral, and cardiovascular diseases. These new compounds have the unique property to inhibit the recognition of several phosphotyrosine containing motifs within all the cellular receptors and cytosolic transducers by a wide spectrum of SH2 domains contained in cytosolic transducers and other effector proteins laying along different pathways of the signal transduction process and with a particularly high affinity for the SH2 domain of the adaptor transducer Grb2, a key element along the pathway to mitogenesis and motogenesis, this last activity leading to invasiveness and to metastasis. The invention also relates to methods for production of the compounds and methods of treatment employing the compounds.
L'invention concerne des peptides et des composés peptidomimétiques représentés par la formule (I), ainsi que des compositions pharmaceutiques les contenant en tant qu'agents pharmacologiques utiles dans la prévention ou le traitement de maladies prolifératives, telles que le cancer, en s'opposant à la croissance de tumeurs et/ou à la métastase tumorale, et le psoriasis et dans la prévention ou le traitement de maladies inflammatoires, allergiques, auto-immunes, virales ou cardio-vasculaires. Ces nouveaux composés possèdent la propriété unique d'inhibition de la reconnaissance de plusieurs motifs contenant phosphotyrosine à l'intérieur de tous les récepteurs cellulaires et des transducteurs cytosoliques par un large spectre de domaines de SH2 contenus dans les transducteurs cytosoliques et dans d'autres protéines effecteurs situées le long de différentes voies du processus de transduction de signal et présentant une affinité particulièrement élevée pour le domaine de SH2 du transducteur adaptateur Grb2, élément clé le long de la voie vers la mitogénèse et la motogénèse, cette dernière activité conduisant au caractère envahissant et à la métastase. L'invention concerne également des procédés de préparation de ces composés et des procédés de traitement mettant lesdits composés en application.
