This invention relates to methods of preventing or reducing the severity of diabetes. In one embodiment, the method involves administering to the individual a peptide having substantially the sequence of a non-conserved region sequence of a T cell receptor present on the surface of T cells mediating diabetes or a fragment thereof, wherein the peptide or fragment is capable of causing an effect on the immune system to regulate the T cells. In particular, the T cell receptor has the V.beta. regional V.beta.6 or V.beta.14. In another embodiment, the method involves gene therapy. The invention also relates to methods of diagnosing diabetes by determining the presence of diabetes predominant T cell receptors.
La présente invention porte sur les méthodes de prévention ou d'atténuation du diabète. Dans une réalisation, la méthode consiste à administrer à l'individu soit un peptide ayant une séquence tirée d'une région non conservée d'un récepteur de lymphocytes T présent à la surface des lymphocytes T médiateurs de diabète, soit un fragment de ce peptide, ce peptide ou fragment de peptide étant capable d'influer sur le système immunitaire pour réguler les lymphocytes T. En particulier, le récepteur de lymphocytes T porte V.beta. régional: V.beta.6 ou V.beta.14. Dans une autre réalisation, la méthode comprend une thérapie génique. L'invention porte aussi sur des techniques diagnostiques du diabète, consistant à déterminer la présence de récepteurs de lymphocytes qui jouent un rôle dominant dans cette maladie.
