Apparatus for performing nucleic acid reactions or manipulations comprises at least one reaction vessel containing a predetermined amount of a reagent for a nucleic acid reaction in a dried state adapted to be reconstituted by addition of aqueous fluid containing a nucleic acid sequence. The reagent preferably comprises a mixture of enzyme and reaction buffer and includes all components of the reaction except the nucleic acid substrate. Reactions and manipulations exemplified include restriction, DNA modification, cDNA synthesis, and dideoxy sequencing.
