Pertactin (formerly 69 kDa protein) is recovered in stable biologically pure form having no detectable adenylate cyclase activity from fermentation broth from the fermentation of Bordetella pertussis as well as from the cells. The broth is processed to selectively remove pertussis toxin (PT) and filamentous haemagglutinin (FHA), the pertactin is precipitated by ammonium sulphate and the precipitate is dissolved in buffer at pH 6.0 to 8.5, the solution then is passed through hydroxyapatite and Q-Sepharose chromatograph columns before final ultrafiltration. Cells are extracted with urea and the extract ultrafiltered and diafiltered. The pertactin is precipitated from the extract and the precipitate processed as above. In a variation, the broth is contacted with ammonium sulphate to precipitate pertactin, PT and FHA, the precipitate is dissolved and the PT and FHA selectively removed, before the solution is passed to the chromatograph columns.
La pertactine (anciennement la protéine de 69 kDa) est récupérée dans une forme stable biologiquement pure n'ayant aucune activité adénylate-cyclase détectable dans le bouillon de fermentation de Bordetella pertussis ni dans les cellules. Le bouillon est traité pour éliminer sélectivement la toxine coquelucheuse (PT) et l'hémagglutinine filamenteuse (FHA), la pertactine est précipitée au sulfate d'ammonium et le précipitat est dissous dans un tampon à pH 6,0 à 8,5, la solution est ensuite passée dans des colonnes chromatographiques d'hydroxyapatite et de Q-Sépharose avant ultrafiltration finale. Les cellules sont extraites à l'urée et l'extrait fait l'objet d'une ultrafiltration et d'une diafiltration. La pertactine est précipitée de l'extrait et le précipitat est traité comme on le décrit ci-dessus. Autre possibilité, le bouillon est mis en contact avec du sulfate d'ammonium pour précipiter la pertactine, la PT et la FHA, le précipitat est dissous et la PT et la FHA sont éliminées sélectivement avant que la solution ne soit passée dans les colonnes chromatographiques.
