The present invention concerns replicons of pestiviruses, in particu-lar replicons of swine fever virus, engineered to have a defective replication there-by having lost infectivity, and further containing a foreign gene. A replicon of the invention contains all the genetic information required for its replication, but lacks essential codons or all codons of at least one of the genes encoding the viral struc-tural proteins E1, E2, E ms or C protein, and consequently cannot generate infec-tious virus particles. Particular replicons are generated with a mutated gene encod-ing a modified N pro protein that no longer controls the cell interferon- induction pathway. Another particular replicon lacks the genes encoding all the structural proteins, the p7 protein and the NS2 protein, and has cytopathogenic properties in transduced cells. The replicons of the invention provide a new vector system that can be used for vaccination, gene delivery and gene therapy applications in mam-mals, including humans, as naked RNA or packaged into any form of delivery ve-hicle.
La présente invention concerne des réplicons de pestivirus, en particulier des réplicons du virus de la fièvre porcine, synthétisés par génie génétique pour avoir une réplication défectueuse, ayant par conséquent perdu de l'infectivité, et contenant en outre un gène étranger. Un réplicon de l'invention contient toutes les informations génétiques requises pour sa réplication, mais présente un manque des codons essentiels ou de tous les codons d'au moins l'un des gènes codant pour les protéines structurales virales E1, E2, Ems ou la protéine C, et par conséquent ne peut pas générer de particules virales infectieuses. Des réplicons particuliers sont générés avec un gène muté codant pour une protéine Npro modifiée qui ne commande plus la voie d'induction d'interféron cellulaire. Un autre réplicon particulier présente un manque des gènes codant pour toutes les protéines structurales, la protéine p7 et la protéine NS2, et a des propriétés cytopathogènes dans des cellules transduites. Les réplicons de l'invention fournissent un nouveau système de vecteur qui peut être utilisé pour des applications de vaccination, d'administration de gène et de thérapie génique dans des mammifères, y compris les êtres humains, sous forme d'ARN nu ou conditionné sous n'importe quelle forme de véhicule d'administration.
