Pharmaceutical formulations are provided that increase the systemic bioavailability of a drug that has low aqueous solubility. The drug is physically entrapped by a spatially stabilized matrix of a hydrophilic polymer, but is not covalently bound thereto. Phospholipid moieties are optionally conjugated to the hydrophilic polymer, and free phospholipids, stabilizing agents and/or other excipients may be incorporated into the formulations as well. Therapeutic methods are also provided, wherein a formulation of the invention is administered to a patient to treat a condition, disorder or disease that is responsive to a particular drug. Generally, administration is oral or parenteral.

Cette invention se rapporte à des formulations pharmaceutiques qui augmentent la biodisponibilité systémique d'un médicament ayant une faible solubilité aqueuse. Le médicament est encapsulé dans une matrice spatialement stabilisée d'un polymère hydrophile, sans être lié à elle par covalence.
