Compounds of formula (I) wherein R is an alkyl, aryl or arylalkyl group wherein the aryl is phenyl, naphthyl or heterocycle; R1 and R2, the same or different, are hydrogen atoms or alkyl groups; R3 is an aryl or arylalkyl group wherein the aryl is as above; R4 is a heterocycle optionally substituted with a heterocycle or phenyl, or it is a phenyl group substituted with a heterocycle (R4 is not imidazole or indole); X is a bond or -OCONH or -CONH- group, processes for their preparation and pharmaceutical compositions thereof, are described. The compounds of formula (I) are endowed with a dual ACE-inhibitory and NEP-inhibitory activity and are useful in the treatment of cardiovascular diseases.
On décrit des composés de la formule (I) dans laquelle R représente un groupe alkyle, aryle ou arylalkyle, où aryle représente phényle, naphtyle ou un hétérocycle, R¿1? et R¿2? sont semblables ou différents et représentent des atomes d'hydrogène ou des groupes alkyle, R¿3? représente un groupe aryle ou arylalkyle où aryle est tel que défini ci-dessus, R¿4? représente un hétérocycle éventuellement substitué par un hétérocycle ou par phényle, ou bien il représente un groupe phényle substitué par un hétérocycle (R¿4? ne peut représenter imidazole ou indole), X représente une liaison ou un groupe -OCONH ou -CONH-. On décrit également des procédés de préparation de ces composés ainsi que des compositions pharmaceutiques les contenant. Les composés de la formule (I) sont dotés d'une double activité d'inhibition, d'une part, de l'enzyme de conversion de l'angiotensine (ACE) et, d'autre part, de l'endopeptidase neutre (NEP), et ils sont utiles dans le traitement de maladies cardio-vasculaires.
