A compound of the formula (I): wherein: A and B together represent an optionally substituted, fused aromatic ring; X and Y are selected from CH and CH, CF and CH, CH and CF and N and CH respectively; R c is selected from H, C1-4 alkyl; and R1 is selected from C1-7 alkyl, C3-20 heterocyclyl and C5-20 aryl, which groups are optionally substituted; or R c and R1 together with the carbon and oxygen atoms to which they are attached form a spiro-C5-7 oxygen-containing heterocyclic group, which is optionally substituted or fused to a C5-7 aromatic ring. These compounds are useful in the preparation of a medicament for inhibiting the activity of PARP for the treatment of : vascular disease; septic shock;ischaemic injury;neurotoxicity;haemorraghic shock;viral infection; or for use as an adjunct in cancer therapy or for potentiating tumour cells for treatment with ionizing radiation or chemotherapeutic agents.

Cette invention concerne un composé de formule (I). Dans la formule (I) selon l'invention, A et B représentent ensemble un cycle aromatique fusionné éventuellement substitué; X et Y sont choisis parmi CH et CH, CF et CH, CH et CF et N et CH, respectivement; Rc est choisi parmi H, un groupe alkyle C1-4; et R1 est choisi parmi un groupe alkyle C1-7, hétérocydyle C3-20 et aryle C5-20, ces groupes étant éventuellement substitués; ou Rc et R1 ensemble avec les atomes de carbone et d'oxygène auxquels ils sont liés forment un groupe spiro-hétérocyclique C5-7 contenant de l'oxygène, qui est éventuellement substitué ou condensé avec un cycle aromatique C5-7. Ces composés sont utiles pour préparer un médicament destiné à inhiber l'activité PARP pour traiter : les maladies vasculaires; le choc septique; l'ischémie; la neurotoxicité; le choc hémorragique; l'infection virale; ou ils peuvent être utilisés en appoints en thérapie anticancéreuse ou pour potentialiser des cellules tumorales en vue d'un traitement avec des rayons ionisants ou des agents chimiothérapiques.
