In a method for making a dry powder formulation for inhalation, a hydrophilic pharmaceutical compound is processed to remove particles having a diameter of less than about 3 microns. A hydrophilic excipient is similarly processed to remove excipient particles having a diameter of less than about 10 microns. The processed pharmaceutical compound and excipient are optionally separately ripened, by exposure to high humidity. They are then blended together to create the formulation, and ripened again, if desired. The removal of fine particles physically stabilizes the formulation. The formulation accordingly provides a consistent respirable dose, even after exposure to high humidity conditions, for long periods of time, such as when a patient opens the packaging of the pharmaceutical, and then uses it over weeks or months.

Selon cette invention, dans un procédé de fabrication d'une formulation de poudre sèche pour inhalation, un composé pharmaceutique hydrophile est soumis à un traitement dans lequel des particules ayant un diamètre inférieur à environ 3 microns sont éliminées. Un excipient hydrophile est soumis au même traitement de façon à éliminer les particules ayant un diamètre inférieur à environ 10 microns. Le composé pharmaceutique et l'excipient, après traitement, sont éventuellement portés à maturation séparément en étant exposés à une forte humidité. Ils sont ensuite mélangés pour obtenir la formulation et à nouveau portés à maturation, si besoin. L'élimination des fines particules stabilise physiquement la formulation. La dose inhalable obtenue à partir de la formulation reste constante sur de longues périodes, même après avoir été exposée à une forte humidité, de sorte que, après avoir ouvert l'emballage du produit pharmaceutique, le patient puisse l'utiliser sur plusieurs semaines ou mois.
