A class of N-substituted piperidine, piperidine and tetrahydropyridine derivatives, further substituted at the 4-position by an optionally substituted alkenyl, alkynyl, aryl-alkyl or heteroaryl-alkyl moiety, are selective agonists of 5-HT1-like receptors, being potent agonists of the human 5-HT1D.alpha. receptor subtype whilst possessing at least a 10-fold selective affinity for the 5-HT1D.alpha. receptor subtype relative to the 5-HT1D.beta. subtype; they are therefore useful in the treatment and/or prevention of clinical conditions, in particular migraine and associated disorders, for which a subtype-selective agonist of 5-HT1D receptors is indicated, whilst eliciting fewer side-effects, notably adverse cardiovascular events, than those associated with non-subtype-selective 5-HT1D receptor agonists.
Une classe de dérivés de la pipérazine, de la pipéridine et de la tétrahydropyridine substitués par un N, substitués également en position 4 par un groupement alcényle facultativement substitué, un groupement alcynyle, un groupement aryl-alkyle ou un groupe caractéristique hétéroaryl-alkyle, sont des agonistes sélectifs de récepteurs homologues du 5-HT1, étant des agonistes puissants du sous-type de récepteur 5-HT1D.alpha. humain, tout en possédant une affinité sélective au moins 10 fois supérieure pour le sous-type de récepteur 5-HT1D.alpha. humain que pour le sous-type de récepteur 5-HT1D.bêta.; ils sont par conséquent utiles dans le traitement et/ou la prévention de conditions cliniques, en particulier de la migraine et des troubles associés, pour lesquels un agoniste sélectif des sous-types du récepteur 5-HT1D est indiqué, tout en provoquant moins d'effets secondaires, notamment des accidents cardio-vasculaires nuisibles, que ceux associés aux agonistes non sélectifs des sous-types du récepteur 5-HT1D.
