The invention provides novel peptides that function in vivo to stimulate insulin release from pancreatic beta cells in a glucose-dependent fashion. These insulin secretagogue peptides are shown to stimulate insulin release in rat islet cells in vitro, and in vivo. The peptides of the present invention provide a new therapy for patients with decreased endogenous insulin secretion, in particular type 2 diabetics. In particular, the invention is a polypeptide selected from a specific group of VIP/PACAP-related polypeptides, or functional equivalents thereof. The invention is also directed to a method of treating a metabolic disease in a mammal comprising administering a therapeutically effective amount of the insulin secretagogue peptides to said mammal. Also disclosed are methods of making the peptides, both recombinant and synthetic.

L'invention concerne de nouveaux peptides fonctionnant in vivo afin de stimuler la diffusion d'insuline par les cellules bêta pancréatiques de manière gluco-dépendante. Ces peptides sécrétrices d'insuline démontrent leur efficacité à stimuler la diffusion d'insuline dans des îlots de Langerhans in vitro, et également in vivo. Les peptides de la présente invention permettent la mise au point d'une nouvelle thérapie pour des patients souffrant d'une baisse de sécrétion d'insuline endogène et en particulier les diabétiques de type 2. Cette invention concerne plus spécialement un polypeptide sélectionné dans un groupe spécifique de polypeptides liés-VIP/PACAP, ou d'équivalents fonctionnels de ces derniers. L'invention concerne également un procédé de traitement d'une affection métabolique chez un mammifère, comportant l'administration d'une quantité thérapeutiquement efficace de peptides sécrétrices d'insuline audit mammifère. Enfin, l'invention concerne des procédés de fabrication de ces peptides, recombinants et synthétiques.
