This invention provides plant arabinogalactan proteins (AGPs) and their genes. AGPs were isolated from Nicotiana alata, Nicotiana plumbaginafolia and Pyrus communis. Amino acid sequences of isolated AGP peptide molecules are presented. Isolated AGP molecules were used to synthesize oligonucleotides probes to prepare, oligonucleotide primers for PCR or prepare RNA probes to screen cDNA libraries of N. alata, N. plumbaginafolia and P. communis. cDNA clones encoding amino acid sequences of isolated AGP molecules were isolated. The invention presents for the first time an intact AGP amino acid sequence derived from a corresponding AGP gene. The instant invention further provides methods useful in obtaining AGP genes encoding an AGP peptide comprising a specific isolated hydroxyproline-rich (OAST-rich) sequence or a specific isolated hydroxyproline-poor sequence.

Cette invention concerne la préparation des protéines végétales arabinogalactanes (PAG) ainsi que celle de leurs gènes. Ces PAGs ont été isolées de Nicotiana alata, Nicotiana plumbaginafolia et de Pyrus communis. On donne la séquence des acides aminés des molécules peptidiques de ces PAGs isolées. Les molécules isolées de PAGs ont été utilisées pour synthétiser des sondes d'oligonucléotides servant à préparer des amorces d'oligonucléotides pour l'amplification enzymatique du génome, ou pour préparer des sondes d'ARN pour examiner des bibliothèques d'ADN complémentaire de N. alata, N. plumbaginafolia et P. communis. On a isolé des clones d'ADN complémentaire codant pour des séquences d'acides aminés de molécules isolées de PAGs. On décrit pour la première fois les séquences intactes d'acides aminés de PAGs dérivées d'un gène correspondant codant pour les PAGs. La présente invention fournit en outre des méthodes utiles pour obtenir des gènes codant pour un peptide de PAG comprenant une séquence spécifique isolée riche en hydroxyproline ou une séquence spécifique isolée pauvre en hydroxyproline.
