A method of plant micropropagation is provided whereby a thin section of non- callus tissue obtained from a monocotyledonous plant, such as sugarcane, wheat or sorghum, is cultured in the presence of a cytokinin and/or an auxin. Optimal regeneration occurs when a basal surface of the thin section is oriented so as to be substantially not in contact with the culture medium. This micropropagation method produces mature monocotyledonous plants by either organogenic or embryogenic regeneration without substantial callus formation. By avoiding use of callus as a starting tissue, this method reduces the likelihood of propagated plants displaying somaclonal variation.

La présente invention concerne un procédé de micropropagation de plantes, consistant à mettre en culture une fine coupe de tissu non calleux provenant d'une plante monocotylédonée, telle que la canne à sucre, le blé ou le sorgho, en présence d'une cytokine et/ou d'une auxine. La régénération optimale se produit lorsqu'une surface basale de la coupe mince est orientée de manière à n'être pratiquement pas en contact avec le milieu de culture. Ce procédé de micropropagation produit des plantes monocotylédonées matures soit par régénération organogène soit par régénération embryogène sans formation de cals. En évitant l'utilisation de tissus calleux comme tissus de départ, ce procédé réduit le risque de variations somaclonales chez les plantes propagées.
