A method of generating genotypic variation in a genome of a plant is disclosed. The method comprising introducing into the plant at least one viral expression vector encoding at least one chimeric nuclease which comprises a DNA binding domain, a nuclease and a localization signal to a DNA-containing organelle, wherein the DNA binding domain mediates specific targeting of the nuclease to the genome of the plant, thereby generating genotypic variation in the genome of the plant.

L'invention concerne un procédé de génération de variation génotypique dans le génome d'une plante. Le procédé comprend l'introduction dans la plante d'au moins un vecteur d'expression viral codant au moins une nucléase chimérique qui comprend un domaine liant l'ADN, une nucléase et un signal de localisation dans une organelle contenant de l'ADN, le domaine liant l'ADN médiant le ciblage spécifique de la nucléase du génome de la plante, en générant ainsi une variation génotypique dans le génome de la plante.
